FPSC Junior Scientific Officer (BS-16) - AFIP
The Armed Forces Institute of Pathology (AFIP) in Rawalpindi, functioning under the Ministry of Defence, is Pakistanβs premier tertiary-care diagnostic and medical research facility. The post of Junior Scientific Officer (BS-16) (Case No. 117/2026) recruits specialized scientific personnel across molecular diagnostics, clinical genomics, and biochemical investigation.
Official Syllabus Specification
The FPSC examination for Case No. 117/2026 consists of a single 100-mark MCQ objective screening test:
Part-I: English (20 Marks)
- Grammar Usage: Tenses, prepositions, subject-verb agreement, and articles.
- Sentence Structuring: Identifying grammatical flaws, modifying phrases, parallel structures, and academic sentence completion.
Part-II: General Science / Specialized Bioscience Test (80 Marks)
Candidates are tested on graduate (BS 4-Year / M.Sc.) curriculum:
- Biochemistry: Amino acids, protein structure hierarchies, enzyme kinetics (Michaelis-Menten kinetics), glycolysis, Krebs cycle, oxidative phosphorylation, lipid metabolism.
- Human Physiology: Homeostasis, blood cell lineages, renal clearance mechanisms, respiratory gas exchange, neuromuscular transmission, and cardiac cycles.

- Mycology: Fungal taxonomy, medically significant fungi (Candida, Aspergillus, Cryptococcus), mycotoxins, diagnostic fungal culture media (SDA), and antifungal modes of action.
- Biological Databases: Primary vs. secondary databases, NCBI Entrez, GenBank accession formats, UniProtKB, Protein Data Bank (PDB), EMBL-EBI.
- Evolutionary Biology & Phylogenetics: Sequence alignment (global vs. local, Needleman-Wunsch, Smith-Waterman), scoring matrices (PAM, BLOSUM62), neighbor-joining and maximum likelihood phylogenetic tree reconstruction.
- Genomics & Transcriptomics: Sanger sequencing principles, High-Throughput / Next-Generation Sequencing (Illumina, Nanopore), RNA isolation, cDNA synthesis, real-time quantitative PCR (RT-qPCR).
- Structural Bioinformatics & Proteomics: Secondary structure prediction (Chou-Fasman, GOR), tertiary structure homology modeling, Ramachandran plot analysis, mass spectrometry (MALDI-TOF, LC-MS).
- Molecular Docking & Drug Discovery: Lead compound identification, structure-based vs. ligand-based drug design, binding energy calculations, hydrogen bonding and electrostatic interactions.
- DNA & RNA Structure: Double helix dimensions (Watson-Crick model), major and minor grooves, A, B, and Z DNA conformations, mRNA, tRNA hairpin loops, rRNA assemblies.
- Chromosomes & Chromatin: Nucleosome core octamer (H2A, H2B, H3, H4), linker histone H1, euchromatin vs. heterochromatin, telomeres, and centromeric repeats.
- Denaturation & Renaturation: DNA melting temperature ($T_m$), factors influencing $T_m$ (GC content, salt concentration), hyperchromic shift, and $C_0t$ kinetic curves.
High-Yield Preparation Topics
- PCR and Primer Design: Melting temperatures, primer dimer formation, annealing temperatures ($T_a = T_m - 5^\circ\text{C}$).
- Enzyme Inhibitors: Distinguishing competitive, non-competitive, and uncompetitive inhibition using Lineweaver-Burk plots.
